분자 요오드(아이오딘)유방 조직에서 바람직한 형태

사람의 유방암 조직에서 NIS 발현 증가에 대한 보고에도 불구하고, 수유 중이 아닌 유방조직은 나쁜 과산화효소로 알려져 있기 때문에, I2는 유방암의 예방과 치료를 위한 요오드 보충제에서 좋은 형태일지도 모른다따라서 요오드 이온의 유기화에 대한 이용률이 적어지게 된다. I2의 우회적인 사용은 과산화효소 활동과  NIS 관여를 위해 필요로 한다놀랍지 않게도 갑상선보다 유선에서 I2를 효과적으로 점유하고 있었으며집중되어 있었다.


생체 내 연구는 I2가 미토콘드리아 막의 선택적인 투과 손실을 일으키고 단백질을 정상적으로 막 내부의 공간에 국한시키는 세포자멸을 유도함으로써, 여러 사람의 암세포에서 증식을 억제시키고 세포자멸을 유도한다는 것을 알아내었다.

I보충제는 양성과 암 모두에서 종양 형성의 크기와 발달을 억제시킨다는 것을 보여주었다.


요오드의 항산화제 효과에 대한 토론에 명시하였듯이요오드는 갑상선 조직 뿐 아니라 갑상선 외 조직에서 항산화제와 항증식 효과를 가진 iodolipid의 생산을 위해 사용되었다I보충제의 항증식 효과가 세포의 과산화를 상당히 감소시키기 때문에, Iodolactone은 유방과 다른 갑상선 외 조직에서 또 다른 보호 메커니즘을 제공할지도 모른다.


또한 최근에 I2는 세포주기 정지와 세포사멸에 관여하는 세포경로를 활성화시키는 아라키돈산과 6-iodolactone에서 파생된 iodolactone 형성을 유도한다는 것을 보여주었다유방암세포는 높은 아라키돈산 농도를 가진다고 알려져 있다이것이 I2가 정상 유방조직이 아닌 유방의 종양세포에만 선택적으로 세포사멸 효과를 가지는 이유이다.


임상의학에서 요오드소금 형태로 사용되는 KI는 이러한 효과를 가지지 않는다.

Molecular Iodine, the Preferred Form in Breast Tissue

Despite reports of the enhanced expression of NIS in human breast cancer tissue, I2 may be the preferred form of iodine supplementation for the prevention and treatment of breast cancer since non-lactating breast tissue is known to be peroxidase-poor and thus is less capable of iodide organification. The use of I2 bypasses the need for NIS involvement and peroxidase activity. Not surprisingly, the mammary gland more effectively captures and concentrates I2 than the thyroid, and more I2 is concentrated in breast than thyroid tissue.


In vitro studies have found that I2 inhibits proliferation and induces apoptosis in some human breast cancer cell lines by causing loss of selective permeability of the mitochondrial membrane, which leads to the release of apoptogenic proteins normally confined to mitochondrial intermembrane space. Supplementation with I2 has been shown to suppress the development and size of both benign and cancerous neoplasias.


As noted above in the discussion of iodine's antioxidant effects, iodine is also used to produce iodolipids with antioxidant and antiproliferative effects in extra-thyroidal tissues as well as in the thyroid gland. Iodolactones may provide yet another protective mechanism in the breast and other extra-thyroidal tissues since the antiproliferative effects of I2 supplementation are accompanied by a significant reduction in cellular lipoperoxidation.


I2 has also recently been shown to induce formation of an iodolactone derived from arachidonic acid, 6-iodolactone (6-IL), which activates cellular pathways involved in cell cycle arrest and apoptosis. Mammary cancer cells are known to contain high concentrations of arachidonic acid, which may help explain why I2 selectively exerts apoptotic effects at lower concentrations only in mammary tumor cells and not in normal mammary tissue.


Of clinical note, potassium iodine, the form used in iodized salt, does not have these effects.
